The Comedy Theatre was built on the site of one of Melbourne's earliest play-houses which was called the old Iron-Pot, which was prefabricated in Manchester by George Selth Coppin in 1855 and it was then left in 1894. The Comedy Theatre was originally a playhouse but during the 1930s and even more during the 1940s when the stage productions were not very common and the Comedy Theatre just screened films.

For fifty years, the Comedy Theatre was built and managed by J.C Williams. The theatre then changed hands in 1978 and it was purchased for $800,000. Today, the theatre is owned and operated by the Mariner Group, one of the leading theatre groups in Australia, which also manages other theatre venues such as the Palace Theatre and the Regent theatre.
